    For example, some dogs chose to leave the testing arena and instead approach the exit door on commencement of each trial Discussion According to the information found in the literature e On completion of the journey, the dog was removed from the car by their handler and led to a room for post-test sampling Dogs were kept behind a barrier on a long line lead with no pressure placed on it by experimenter 1 whilst experimenter 2 placed the bowl in one of the five locations Latency s , measured live with a stopwatch, was recorded for the time it took for the dog to travel from the starting point at the barrier to reaching the bowl, with a maximum of 30 s allowed before being called back behind the barrier to begin the next trial Unlike many pet dogs, this particular population do not routinely undergo car travel, so the novelty of the event may have caused a more pronounced stress response The criteria for selection were: age between 1 and 10 years estimated by standard veterinary methods ; physically healthy; presence of behavioural disorders detected by the veterinarian ; permanence in the shelter for at least 9 months Table 1 Although the association between epilepsy and the ECS has not been fully elucidated, the complex relationship between brain excitability and the ECS suggests that phytocannabinoids may induce beneficial effects on epilepsy, paving the way for the possibility of developing new treatments involving the use of compounds, especially CBD, that selectively target individual elements of the endocannabinoid signaling system There were no significant effects of treatment on the combined stress measure at either timepoint, or on the change from baseline to test Overall, it was evident that some measures of canine stress changed significantly from baseline after exposure to these specific events, suggesting the situations elicited a negative emotional response in dogs .

    Indeed, the dogs often sat by the door, possibly awaiting their caregiver's return Dogs with idiopathic epilepsy seizures with no other abnormalities found; presumed genetic basis were enrolled in this study However, no significant pharmacokinetic interactions were found between CBD and phenobarbital when simultaneously administered to healthy dogs

    There are very few papers on the effect of different treatments in this behavioural category 68 , for example, did not find an effect of the appeasing pheromone in reducing displacement activities in shelter dogs

    In the last few years, several pharmacokinetic studies on CBD were conducted in dogs 25 — On every third trial, one of three noises was played 0 Some psychoactive medications, including herbal supplements or pheromonal products, have been used to lower the level of anxiety of dogs e Due to the ever-changing human-pet relationship, there is an increasing requirement for dogs to fit into human lifestyles which can cause stress and anxiety to both people and their pets Figure 2 Schematic of the population and sub-population of dogs recruited to the study, and number of dogs included in analyses for each behavioural task However, as epilepsy is a heterogenous disease, even within-breed, it is possible that a larger sample is needed Testing was ceased for any dogs that appeared fatigued by the task e The impact of epilepsy and its comorbidities on affective state are yet to be explored in the dog in an empirical manner, with studies to date relying on survey-based owner reports In a previous randomised placebo-controlled crossover trial of an MCT-enriched diet, dogs with epilepsy were found to have reduced stranger-directed fear during the MCT phase than the placebo phase 19 , with anxiolytic effects of MCTs also seen in rodents At the time of enrollment, all patients were having multiple seizures per month despite receiving standard anti-convulsant medications Prior to the study, one dog was identified as having mobility concerns and so was unable to safely enter the car With and without CBD, the oil administration did not require any kind of particular interaction since the oil was mixed with some meat; in any case, due to their behavioural disorders, most of the dogs did not allow any form of interaction with humans There are several hypotheses, but none have been verified .

    The value of emissivity was set at 1 as per manufacturer guidelines, replicating previous studies 44 , Albeit smaller reductions, there was a statistically significant larger median reduction of seizures in the CBD group and a larger number of dogs in the CBD group showing a reduction in seizures compared to placebo By contrast, the activation of CB1 receptors mediates the anticompulsive effects, the enhancement of fear extinction, the blockade of reconsolidation and the ability to prevent the long-term anxiety-producing consequences of stress The study was a clinical trial and its purpose was to determine if CBD treatment can decrease disturbed and stressed behaviour in shelter dogs, in terms of decrease in displacing activities, stereotyped and aggressive behaviour All dogs were housed in pairs within kennels at the Waltham Petcare Science Institute Leicestershire, UK with free access to indoor and outdoor environments The hypothesis that the ECS plays a role in the modulation of emotional behavior is supported by the demonstration that CB1 receptors and Fatty Acid Amide hydrolase FAAH - the enzyme responsible for the degradation of endocannabinoids , as well as the endocannabinoids AEA and 2-AG, are expressed and produced in brain areas such as the amygdala, nucleus accumbens, hippocampus and prefrontal cortex involved in stress, fear, emotions and reward mechanisms If a dog was distressed at any point during the study, the tasks were abandoned, and the dog returned to their owner The parameters age, sex, breed, and housing location were considered when balancing the groups Dogs in the separation group were habituated to the testing room until they were deemed comfortable in the environment with their experienced handler present In fact, following rectal administration of a suppository containing mg of CBD, corresponding to a dose between 6 Endogenous cannabinoids produce antinociceptive and antihyperalgesic effects at peripheral, spinal and supraspinal levels Finally, all the measures were combined to create an overall stress measure using a factor analysis Despite the relative frequency of people transporting dogs and the welfare implications this may have on pets and humans, the transport of companion animals has not been widely researched Although mild stress is not considered harmful 9 , welfare is compromised if an animal experiences high levels of acute stress or is exposed to prolonged stressors 10 which can lead to chronic physical and emotional health issues In trying to minimize the number of variables that could have affected the results in such a variable environment, we chose dogs that had been in shelter for at least nine months and displayed signs of chronic stress .

    Dogs under treatment displayed the same level of attention towards the environment before and after the treatment A major reason for loss of data in the judgement bias task was dogs with IE being unable to learn the discrimination within the allotted trials Most papers published on cannabinoids in pruritus deal with ALIAmides i Attention bias tasks are based on the principle that anxious individuals are more likely to direct attention to potentially threatening, or even innocuous, stimuli 21 , 30 , Briefly, ellipses were drawn around the anterior surface region of the eye, the exterior surface of the ear flap and the anterior surface of the nose The authors further hypothesized that attentional biases related to threat in patients with TLE may exacerbate disease, by sustaining emotional vulnerability and seizure occurrence Consistent with these observations, studies examining canine stress have described cortisol levels increasing 58 , 59 and HRV lowering in response to a perceived negative stimulus 60 , 61 , with dog behaviors related to lip licking, such as hypersalivation, barking and restlessness occurring more frequently during car travel 4 Eligibility was screened using an online sign-up questionnaire

    There are reports of liver enzyme elevations in humans as well and periodic bloodwork monitoring is recommended for all patients taking CBD for this reason

    These are often unavoidable and are commonly related to events such as car travel 3 , 4 and separation from caregivers or conspecifics 5 — 8 Attention bias tasks have been tested in a variety of species, and recently validated in sheep 42 , 43 and beef cattle 44 by pharmacologically inducing anxiolytic, anxiogenic, and controlled states prior to testing and quantifying behavioural measures such as attention paid to the threat and latency to feed .

    Terms included in the Qualitative Behavioral Assessments QBA used to measure dog behavior during baseline, the separation, and car travel tests With specific regard to the canine species, the presence of cannabinoid receptors or their ligands has been identified in skin and skin appendages of healthy dogs and dogs with atopic dermatitis AD 4 — 8 , gastrointestinal tract 9 , 10 , peripheral and central nervous system 11 — 13 , joints 14 and embryo Moreover, PEA also acts indirectly by elevating the levels, reducing the degradation, and increasing the affinity of endocannabinoids for their receptors Detailed results for the analyses of individual terms from the QBA are presented in the Supplementary material Sixteen dogs successfully completed the study Three models were run for each measure, with analyses looking separately at car and separation stress tests, as well as at both stress tests combined However, data in humans suggest that these neurological signs in the dogs are attributable to effects of THC and not CBD A more recent study investigated the efficacy a lower 1 In contrast, in a recent study of dogs trained on the same spatial discrimination, only one dog 0 Dogs had experienced a mean of 3 .

    CBD has the potential to alter the blood concentration of anticonvulsants in humans Additionally, some measures of stress in dogs were significantly affected following administration of CBD, suggesting it may have efficacy as an intervention for acute stress in dogs In mammals, the ECS is very complex and modulates different kind of organism responses A pharmacokinetic study was completed at CSU and published in Throughout this habituation period and during all behavioural tasks, each dog was monitored for potential signs of stress, anxiety and frustration caused by the setting or the tasks potential behaviours listed in Supplementary File 1 Full size table The selected dogs showed severe behavioural disorders such as compulsively licking the cage walls, chewing on objects until they were destroyed, coprophagy or having attacks of aggression such as to lead to self-injury; none were under therapeutic, pharmacological or behavioural treatment If the true mechanism of seizure control were to be established, this may help us understand which patients would benefit from its use All methods were performed in accordance with the relevant guidelines and regulations The endocannabinoidome: the world of endocannabinoids and related mediators These were each presented only once to avoid learning effects, and were presented in the same order MID, NP, NN separated by four pseudo-randomised trials two P and two N The absence of a history of cutaneous or systemic disease, the histopathological findings, and the remission after symptomatic treatment and discontinuation of the CBD product allowed the Authors to consider a possible CADR cutaneous adverse reaction to drugs to CBD, as described in men 96 , or to additional substances in the vehicle As it is well known 43 , 58 , sheltered dogs in general and the dogs in this study in particular, suffer from inter- and intra-specific social deprivation, total lack of interactions at night, and lack of exercise because they are in cages People with epilepsy are more likely to report anxiety than those without epilepsy 25 , or patients with other chronic medical issues Different oral formulations are likely to reduce the incidence of these signs Stress is a mental, physiological, or emotional state characterized by a factor that is altering the homeostasis of a living organism Epilepsy In recent years, phytocannabinoids have been emphasized in treating various neurological disorders, including epilepsy Cortisol also significantly increased for the CBD group for the car test Some authors suggested that displacement activities are behavioural constituents of the adaptive stress response 67 ; morphologically, in nonhuman primates these behavioural patterns have something to do with body care: self-grooming, scratching, body shaking, stretching and yawning .

    Further, it is widely available in a range of products claiming positive benefits, stress-reducing properties and improved pet emotional wellbeing A Separation test room setup showing position of vet bedding center, with a dog on top , then clockwise from top: cardboard box, water bowl, dog bed, crate, and toys Any missing data were imputed using multiple imputation Prevalence and risk factors for canine epilepsy of unknown origin in the UK In humans, an antipsychotic activity of CBD was assessed and found to reduce the occurrence of apomorphine-induced stereotypies 64 , but the mechanism by which CBD exerts its anxiolytic effects has not been fully clarified, yet A total of videos were randomly assigned between the three raters Further work will be required to fully understand the relationship between stress and activity in canines, and the impact CBD has on it Canine keratinocytes also express TRPV1 receptors 6 It is therefore possible that surface body temperature may not be suitable as a standardized measure of canine stress unless under highly controlled conditions

    Dog behavior Dog behavioral data from baseline and during the two testing sessions were collected from video footage by trained observers

    Exposure to a separation event and car travel Pet dogs routinely experience being left alone, as well as traveling in a vehicle, although the absolute and relative impacts that these events have on both physiological and behavioral measures of canine stress are not well-understood Thus, in this species, it is not even clear which behavioural patterns can be considered displacing activities that, in turn, are behavioural components of the adaptive stress response, probably causing anxiolytic effects This review aims to summarize all data published so far about the pharmacokinetics, efficacy, and tolerability of Cannabis derivatives, specifically CBD and cannabidiolic acid CBDA , in the canine species Collectively these characteristics are suggestive of a more relaxed emotional state in CBD treated dogs Similarly, sex was observed to affect metabolism of some drugs To date, the authors are not aware of attention bias paradigms that have been developed with dogs as the test subject rather than the threatening stimuli, but such methods potentially offer advantages over judgement bias tasks, which often involve lengthy training periods and lead to attrition of subjects All dogs from case and control groups were eligible if they were over one year of age, had no health problems affecting vision, hearing or mobility, and did not show marked negative behavioural responses to unfamiliar people or around food bowls, as these factors may influence ability to perform the tasks included in the test The crate was fixed in place with bungee cords and contained a single piece of non-slip vet bedding Figure 2B Little is known about the veterinary use of CBD in dogs Indeed, somnolence, loss of appetite and diarrhea have been reported as common signs in human clinical trials during treatment with CBD 86 , .

    According to these considerations, the results presented here acquire value since they suggest a possibility of response to the treatment in a challenging environment, that could be even greater in an environment where the limitations described above are less present and the possibilities to control the dogs are greater This helps us extrapolate a dose to use during clinical trials to study its effect on various diseases There were no significant changes from baseline to post-test for IgA or glucose Figure 3 Full size image To compare responses of IE and control dogs to the ambiguous probe locations in the test phase, unadjusted latency data were first used in a linear mixed model LMM The sound source was hidden by the experimenter standing behind the MID bowl What are CSU veterinarians studying? Researchers are evaluating the pharmacokinetics how much of a medication is absorbed and how long it is detectable in the bloodstream and safety of CBD in dogs and cats Imepitoin has been found to have anxiolytic as well as antiseizure effects in physically healthy dogs 67 , 68 , 69 ; however, no changes in anxiety levels were reported by owners of dogs treated with imepitoin for IE If dogs exhibited any signs of distress or sensitisation Supplementary Table S1 , the sound was stopped and the attention bias task was not undertaken The study consisted of a parallel design, so individual differences in dog behavior and preferences may have contributed additional variation For these dogs, the P bowl location was baited five times before moving straight into the attention bias task trials to reinforce this location Any product that may be evaluated in this article, or claim that may be made by its manufacturer, is not guaranteed or endorsed by the publisher Factor scores were then modeled as above using linear mixed effects models, to determine the effect of treatment and timepoint Despite some non-significant results, our findings suggest that it is worth doing more research to further investigate the effect of CBD on dog behaviour; this would be certainly valuable because the potential for improving the welfare of dogs in shelters is priceless CBD has low acute intravenous i From obtained results, some evidence exists supporting the beneficial role of CBD Studies on clinical efficacy of CBD-based products in the treatment of skin diseases in dogs Following the recent COVID pandemic, the incidence of separation-related behaviors in dogs has increased and cases are expected to continue to rise 12 , 17 , No dogs showed disease symptoms during the study, except for one dog Gargamello, under treatment that had a single episode of diarrhoea, during the second day of T2, which disappeared without pharmacological intervention; so we did not exclude this dog from the study .

    The placebo consisted of extra virgin olive oil only

    Furthermore, no experiments to investigate the neurobiological correlates of displacement activities and their relationships with negative emotional states have ever been carried out in the domestic dog Dog owners are often left feeling frustrated due to the damage pets cause to their property, as well as not being able to successfully manage the problem behavior While the second one is responsible for the psychotropic and toxic effect, both in humans and animals e Rather than an intermediate response to the middle bowl as seen in Cockburn et al Aaron Rozental and Dr .

    The mechanism by which CBD exerts its anticonvulsant effects is unknown Prior to the study, dogs were habituated to the testing environments and associated equipment and underwent appropriate training to facilitate sample collections e In this study a more notable effect was observed between various baseline measures and exposure to car travel, compared to baseline and separation test Table 1 resumes the data obtained from the pharmacokinetic studies published so far The gastrointestinal absorption of CBD seems very low Overall, these data provide a more detailed understanding on how dogs cope during these routine events Once separated from their owners, all dogs were given a 10—min habituation period in the study room with the experimenters before the behaviour tasks began Despite a certain number of available pharmacokinetic studies, the kinetic profile of CBD has yet to be fully known, probably because of the very different experimental conditions Serum cortisol and heart rate, both widely used physiological measures of mammalian stress, were elevated significantly following exposure to the car test Factors extracted by the factor analysis of all measures when analyzed with both stress tests combined, or with separation and car tests separately Indeed, although several studies on the pharmacokinetics of CBD in dogs have been conducted, the kinetic profile of CBD is not yet fully known, probably because of the very different experimental conditions used, such as different oily vehicles sunflower lecithin, MCT, and sesame oil , pharmaceutical forms tablets, chews, microencapsulated oil beads, or drops , type of CBD synthetic and purified or full spectrum extract and route of administration oral, rectal, intranasal or oral transmucosal Table 1 In both studies, phytocannabinoids [0 Although future studies could disprove this result, it is possible to hypothesize that CBD is effective in chronic but not in acute pain Dogs in the present study received a larger oral dose of CBD via capsules that was administered more acutely, just 2 h prior to testing However, studies were prevalently focused on short- to medium-term treatment, while CBD is usually employed for long-term treatment .
